چکیده

Abstract Background Plasmodium falciparum parasite populations in Ethiopia have been experiencing local selective pressures from drugs and immunity, leading to evolutionary adaptation. However, there was a paucity of data on genomic characterization adaptations P. isolates the central area Ethiopia. Methods Whole-genome analysis 25 Ethiopia, specifically West Arsi, were studied determine their genetic diversity, population structures, signatures selection known drug resistance alleles against global Cambodia, Thailand, DR Congo, Malawi. Results A total 18,517 high-quality single-nucleotide polymorphisms (SNPs) identified Ethiopian isolates. About 84% had F WS value > 0.95 showing dominant single genotype infection most at time collection with little potential for out-crossing as expected areas low transmission intensity. Within-host diversity infections significantly different East African (p < 0.001), but not Southeast Asian (P 0.05). significant structure has observed by PCA differentiation between parasites (Fst ~ 10%) 18%), suggesting limited gene flow independent evolution population. Moreover, 125 genes under balancing found that include ama1 , trap eba175 lsa3 previously targets human host immunity. Recent directional using integrated standardized haplotype score (IHS) did detect any Pfcrt Pfdhfr Pfdhps Pfmdr 1, PfK13 genes. resistance-conferring mutations showed least one SNP marker fixed these genes, . Conclusion region structurally diverged both other populations. Malaria within-host carry chloroquine markers despite withdrawal this treatment

Comparative Transcriptional and Genomic Analysis of Plasmodium falciparum Field Isolates

Mechanisms for differential regulation of gene expression may underlie much of the phenotypic variation and adaptability of malaria parasites. Here we describe transcriptional variation among culture-adapted field isolates of Plasmodium falciparum, the species responsible for most malarial disease. It was found that genes coding for parasite protein export into the red cell cytosol and onto its...

RESISTANCE OF PLASMODIUM FALCIPARUM TO CHLOROQUINE IN SOUTH EASTERN IRAN

In vivo and in vitro assessments of the response of P. falciparum to chloroquine using WHO standard kits and techniques were carried out in I ran Shahr, Sistan and Baluchestan province of Iran in 1985. In the in vivo assessment, 24 malaria patients treated with chloroquine (25mg/kg over three days) were followed up for one to four weeks. The mean parasite clearance time was 4.3 days and in...

Plasmodium falciparum Transcriptome Analysis Reveals Pregnancy Malaria Associated Gene Expression

BACKGROUND Pregnancy-associated malaria (PAM) causing maternal anemia and low birth weight is among the multiple manifestations of Plasmodium falciparum malaria. Infected erythrocytes (iEs) can acquire various adhesive properties that mediate the clinical severity of malaria. Recent advances on the molecular basis of virulence and immune evasion have helped identify var2csa as a PAM-specific va...

Determining the severity of Plasmodium falciparum malaria in Ethiopia.

BACKGROUND In the majority of health centers in Ethiopia, the severity of falciparum malaria has been determined by parasitemia alone. However, it has been suggested that the use of peripheral infected RBC counts as an indicator of disease severity by itself is insufficient. Therefore, this study was performed to assess the severity of falciparum malaria infection in three Ethiopian localities ...

Independent Origin of Plasmodium falciparum Antifolate Super-Resistance, Uganda, Tanzania, and Ethiopia

Super-resistant Plasmodium falciparum threatens the effectiveness of sulfadoxine-pyrimethamine in intermittent preventive treatment for malaria during pregnancy. It is characterized by the A581G Pfdhps mutation on a background of the double-mutant Pfdhps and the triple-mutant Pfdhfr. Using samples collected during 2004-2008, we investigated the evolutionary origin of the A581G mutation by chara...
